The church was built as part of the expansion of the castle complex under Duke (later Grand Duke) Carl II of Mecklenburg-Strelitz (1741–1816). Hohenzieritz Castle served him as a summer residence and was developed into a prestigious estate from the 1770s onwards, including an English landscape park (one of the oldest on the continent).

The organ was built by Barnim Grüneberg in 1892 and restored in 2014, making it an important instrument in the region. Prillwitz is picturesquely situated in the Tollense Basin of the Mecklenburg Lake District.
