In 1000, Emperor Otto III donated the game reserve around Colmberg to the Bishopric of Würzburg. In 1128, Colmberg became a fief of the Counts of Hohenlohe. In the period from 1150 to 1240, the old fortification was expanded into the Imperial Castle of Colmberg under the German emperors and their governors. In 1254, Count Friedrich von Truhendingen seized the imperial fief of Colmberg after the Staufen emperors died out. Counts Friedrich VIII and Konrad von Truhendingen sold Colmberg to Burgrave Friedrich IV of Nuremberg for 6,200 pounds of Haller Pfennig. Around 1320, Colmberg was the seat of a burgrave (later margrave) upper office with a bailiff and treasurer. The settlement of "Altenstatt" near Colmberg was first mentioned in 1361. The parish church of St. Ursula was first mentioned in 1375. In addition to the parish church, there was the chapel of Colmberg Castle, as a baptismal font from the late 13th century shows; in 1451, Margrave Albrecht Achilles had the castle chapel rebuilt on the first floor of the bower building and appointed a chaplain who was entitled to two thirds of the Colmberg tithe.
